| I. Background | |
| ADAM9（A Disintegrin and Metalloprotease 9）基因定位于人类染色体 8p11.22，编码 一种多结构域Ⅰ型跨膜蛋白，属于 ADAM（去整合素金属蛋白酶）家族成员。其蛋白产物 分子量为 40-45 kDa，广泛表达于单核细胞、巨噬细胞及多种肿瘤细胞中，但在正常组织中 表达有限。在功能方面，ADAM9 顺利获得裂解 TEK、KDR 等蛋白参与肿瘤发生与血管生成， 并顺利获得整合素相互作用调节细胞运动。此外，它还可能作为淀粉样前体蛋白的α分泌酶，与 神经退行性疾病相关。ADAM9 还展现出非蛋白酶依赖功能，如介导细胞粘附并促进癌症 转移。 ADAM9 基因顺利获得多结构域介导肿瘤微环境重塑、神经代谢调控及炎症应答，其异常 表达与多种病理过程密切相关。随着 ADC 药物及 microRNA 疗法的推进，ADAM9 已成为 癌症精准治疗的重要靶点，未来需结合多组学技术深入解析其功能网络，有助于临床转化。 |
